چکیده
In this study, the complete cDNA sequence (1552 bp) of cold-inducible RNA binding protein gene (cirbp gene) was successfully cloned from liver in Acanthopagrus schlegelii (initial weight: 15.0 ± 2.3 g). Results showed that Ascirbp A. schlegelii) has 24 phosphorylation sites, no signal peptide, and transmembrane helix structure. AsCIRBP, with a molecular weight 18.84 ku an isoelectric point 9.04 stable encodes 182 amino acids. Subcellular localization analysis it located nucleus. Sequence alignment results AsCIRBP acid sequences various fishes including black porgy were highly conserved, especially recognition motif (RRM). Those real-time quantitative PCR (qRT-PCR) demonstrated specifically expressed tissue its expression significantly increased under cold stress or acclimation. The interference experiment Ascirbp-dsRNA could suppress through intraperitoneal injection. After silencing gene, RNAi groups more severely damaged structure prone to apoptosis than control groups. study on linkage between mitochondrial pathways changes had significant effect key genes apoptosis. most striking result expressions bcl-2 apaf-1 decreased significantly, which can block normal apoptotic process. disruption process, p53, bax, cyto-c, caspase-9, caspase-3, diablo, caspase-1 affected. These suggest inhibit protect at low temperatures.

Temperature regulates splicing efficiency of the cold-inducible RNA-binding protein gene Cirbp.
In mammals, body temperature fluctuates diurnally around a mean value of 36°C-37°C. Despite the small differences between minimal and maximal values, body temperature rhythms can drive robust cycles in gene expression in cultured cells and, likely, animals. Here we studied the mechanisms responsible for the temperature-dependent expression of cold-inducible RNA-binding protein (CIRBP). In NIH3T...

A gene (htrA) coding for a stress-inducible HtrA-like protein from Lactobacillus helveticus CNRZ32 was cloned, sequenced, and characterized. The deduced amino acid sequence of the gene exhibited 30% identity with the HtrA protein from Escherichia coli; the putative catalytic triad and a PDZ domain that characterize the HtrA family of known bacterial serine proteases were also found in the seque...
متن کاملCold-inducible RNA-binding protein CIRP/hnRNP A18 regulates telomerase activity in a temperature-dependent manner
The telomerase is responsible for adding telomeric repeats to chromosomal ends and consists of the reverse transcriptase TERT and the RNA subunit TERC. The expression and activity of the telomerase are tightly regulated, and aberrant activation of the telomerase has been observed in >85% of human cancers. To better understand telomerase regulation, we performed immunoprecipitations coupled with...

متن کاملA Scallop IGF Binding Protein Gene: Molecular Characterization and Association of Variants with Growth Traits
BACKGROUND Scallops represent economically important aquaculture shellfish. The identification of genes and genetic variants related to scallop growth could benefit high-yielding scallop breeding. The insulin-like growth factor (IGF) system is essential for growth and development, with IGF binding proteins (IGFBPs) serving as the major regulators of IGF actions. Although an effect of IGF on gro...
